How Long Can Psilocybin’s Antidepressant Effects Last?
🍄 Trial Participants Got a Measured Dose. Mushrooms Don't Come Labeled.
Psilocybin content can vary between species, batches, and even parts of the same mushroom. What you buy isn't always what you get—estimate potency before you assume.

The clinical trial included 35 adults with major depressive disorder. Participants received either 25 milligrams of psilocybin or 100 milligrams of niacin, which served as the active placebo.
Depression scores improved significantly by the eighth day after psilocybin. Participants also reported benefits on several secondary measures for more than three months.
By the one-year follow-up, however, the psilocybin and placebo groups were no longer significantly different on clinician-rated depression scores.
In This Article:
The Study at a Glance
- Participants: 35 adults with major depressive disorder (17 psilocybin, 18 niacin)
- Doses: A single 25 mg dose of psilocybin vs. 100 mg of niacin as an active placebo
- Day 2: Early signs of improvement on self-reported measures
- Day 8: Significant reduction in clinician-rated depression scores
- 3+ months: Self-reported benefits on several secondary measures
- 1 year: No statistically significant difference from placebo on clinician-rated scores
- Published: JAMA Network Open, May 2026 (Karolinska Institutet, Sweden)
Psilocybin Produced a Rapid Effect
The biggest difference appeared shortly after the treatment session.
Participants receiving psilocybin showed significant reductions in depression symptoms by day eight, with evidence of improvement beginning as early as day two. Self-reported benefits on some secondary outcomes lasted longer than the primary clinician-rated difference.
That matters because conventional antidepressants can take weeks to produce their full effect. A treatment capable of helping within days could eventually fill an important gap, but rapid improvement and durable recovery are different outcomes.
By one year, clinician ratings no longer showed a statistically significant advantage for psilocybin over niacin. That does not prove that the treatment stopped helping every participant. It means the researchers could no longer confidently attribute the difference between the groups to psilocybin.
The study was also small and was not designed to draw firm conclusions from every long-term secondary outcome.
⚠️ Keep It in Context: With only 35 participants, this trial could reliably detect only large differences between groups. "No longer significant" at one year is not the same as "the effect disappeared"—it means the evidence was no longer strong enough to separate psilocybin from placebo.
A Supported Session Is Not Just a Dose
Participants did not simply receive psilocybin and go home. The protocol included:
- Screening
- Preparation
- A controlled dose
- Professional support during the experience
- Follow-up
Those conditions are part of the treatment researchers are studying.
The trial also documented meaningful risks. Two participants required additional care for prolonged anxiety after their psilocybin sessions. Psychedelic experiences can be psychologically difficult even when the compound and dose are known, and professional support is present.
🚨 Safety Note: These results should not be interpreted as evidence that someone can recreate psilocybin therapy through unsupervised use.
Mushroom Potency Cannot Be Judged by Appearance
Clinical researchers administered a measured 25-milligram dose of identified psilocybin. Mushroom material presents a very different situation.
Psilocybin and psilocin concentrations can vary between:
- Species
- Batches
- Individual mushrooms
- Different portions of the same mushroom
A strain name, photograph or previous experience cannot establish the potency of a new sample.
A Psilocybin QTest answers one specific question at home: How potent is the mushroom sample being tested?
The QTest provides an estimate of potency when used with a properly prepared, representative sample. It does not identify every compound present, detect every possible contaminant or determine whether the rest of a batch has the same potency.
For broader information, laboratory testing can analyze an accepted mushroom sample for psilocybin, psilocin and other natural or synthetic tryptamines.
Two Tools, Two Different Questions
- At-home Psilocybin QTest: How potent is this mushroom sample? (Get the Psilocybin QTest)
- Laboratory analysis: Which natural or synthetic tryptamines are present in the submitted sample? (Submit a Sample)
At-home potency testing and laboratory analysis answer different questions. Neither result should be treated as providing every piece of information about an entire mushroom batch.

What you buy isn't always what you get. Laboratory testing can screen an eligible mushroom, plant, or extract sample for psilocybin, psilocin, and other tryptamines.

Drug checking cannot recreate clinical screening, preparation, supervision, or mental health support. It also cannot predict whether someone will experience relief, anxiety, or another psychological response.
What it can do is replace some assumptions with information.
Transparency Testing offers Natural and Synthetic Tryptamine Panels for eligible mushroom, plant, and extract samples. These panels can screen for compounds including:
- Psilocybin
- Psilocin
- N,N-DMT
- 5-MeO-DMT
- Several synthetic tryptamines
⚠️ Important: A laboratory result only describes the submitted sample. It does not establish that the rest of a batch is identical or that consuming it will be safe.
What the One-Year Results Mean
This study strengthens the evidence that supported psilocybin therapy can produce rapid reductions in depression symptoms. It does not establish that one session reliably produces a year of relief.
Longer and larger trials will need to determine whether additional treatment, ongoing therapy, or carefully timed follow-up sessions can extend the initial response.

Frequently Asked Questions
How long do psilocybin's antidepressant effects last?
In this trial, a single supported 25 mg dose significantly reduced clinician-rated depression scores by day eight, and self-reported benefits on several secondary measures lasted more than three months. By one year, the difference between the psilocybin and placebo groups was no longer statistically significant.
Why was niacin used as the placebo?
Niacin served as an active placebo, meaning it can produce noticeable physical sensations. Researchers use active placebos to make it harder for participants to tell which treatment they received.
Can I recreate psilocybin therapy on my own?
No. The trial included screening, preparation, a controlled dose, professional support during the experience, and follow-up. Even with that support, two participants required additional care for prolonged anxiety.
Can I tell how potent mushrooms are by looking at them?
No. Psilocybin and psilocin concentrations vary between species, batches, individual mushrooms, and different parts of the same mushroom. A Psilocybin QTest can estimate the potency of a properly prepared, representative sample.
What's the difference between the Psilocybin QTest and lab testing?
The QTest estimates potency at home. Laboratory testing can analyze an accepted sample for psilocybin, psilocin, and other natural or synthetic tryptamines. Neither result describes an entire batch.
